Biological pathway information for Coagulation from PathBank. Blood coagulation can be initiated by either an extrinsic or an intrinsic pathway, resulting in a cascade of serine protease activation that ultimately leads to the formation of thrombin, which converts soluble fibrinogen to an insoluble fibrin clot.

– Tissue factor pathway inhibitor - TFPI. The extrinsic pathway of blood coagulation Upon the introduction of cells, particularly crushed or injured tissue, blood coagulation is activated and a fibrin clot is rapidly formed. The protein on the surface of cells that is responsible for the initiation of blood clotting is known as tissue factor, or tissue thromboplastin. The traditional view of the regulation of blood coagulation is that the initiation phase is triggered by the extrinsic pathway, whereas amplification requires the intrinsic pathway. The extrinsic pathway consists of the transmembrane receptor tissue factor (TF) and plasma factor VII/VIIa (FVII/FVIIa), and the intrinsic pathway consists of plasma FXI, FIX, and FVIII. Extrinsic Pathway The extrinsic pathway is activated by external trauma that causes blood to escape from the vascular system.

These reactions are initiated by tissue injury and result in the formation of a fibrin clot (Figure 1). Figure 1: The blood coagulation cascade.

Biochem a chain of reactions associated with a Explanation of extrinsic pathway of coagulation Title: Coagulation Cascade Pathway Author: Sigma-Aldrich Co. LLC Subject: Coagulation Cascade Keywords: Coagulation Cascade Pathway, intrinsic pathway, extrinsic pathway The extrinsic pathway is initiated upon vascular injury which leads to exposure of tissue factor, TF (also identified as factor III), a subendothelial cell-surface glycoprotein that binds phospholipid. The green dotted arrow represents a point of cross-over between the extrinsic and intrinsic pathways.

Extrinsic Pathway The extrinsic pathway is activated by external trauma that causes blood to escape from the vascular system. This pathway is quicker than the intrinsic pathway. It involves factor VII. Intrinsic Pathway The intrinsic pathway is activated by trauma inside the vascular system, and is activated by platelets, exposed endothelium, chemicals, or collagen. This pathway is slower than the extrinsic pathway, but more important.
